چکیده

Streptomyces antibioticus producing antimycin A and actinomycin did not fit in cell-wall types of aerobic actinomycetes. In addition to universal actinomycetes cell-wall components, i. e. glucosamine, muramic acid, glutamic acid and alanine, diaminopimelic acid (DAP), lysine, leucine (isoleucine), glycine, arabinose, glucose, galactose and mannose were detected as major constituents. Phase of the life-cycle influenced the distribution of serine, aspartic acid, lysine and DAP isomers. LL-DAP was found rather than meso form. MesoDAP was consistently revealed after 48 hr incubation and not at other times, and as distinct from LL was not strict alternative with lysine. The occurrence of meso-DAP, relatively small amounts of murein glucosamine (2.2-5.5 %) and lysozyme resistance suggest a similarity to nocardiae cell-walls. Decrease of murein hexosamine was unfavourable for antimycin A yields. Variation of the murein amino acids stimulated production of actinomycin.

The possible involvement of a plasmid(s) in actinomycin synthesis by Streptomyces parvulus and Streptomyces antibioticus.

The treatment of Streptomyces parvulus and Streptomyces antibioticus with acriflavine or novobiocin resulted in the loss of ability to produce actinomycin. The concomitant loss of ability to form aerial mycelium and the incidence of auxotrophic progeny (S. parvulus) were both low relative to the loss of the antibiotic-producing property. Protoplast fusion induced by polyethylene glycol 4000, us...

Sensitivity of transcription by purified Streptomyces antibioticus RNA polymerase to actinomycin.

DNA-dependent RNA polymerase has been purified 400-fold from 48 hr old, actinomycin producing cells of Streptomyces antibioticus. Sodium dodecyl sulfate polyacrylamide gel electrophoresis of the purified 5. antibioticus polymerase reveals the presence of bands corresponding to the 8, 8' and Q subunits of Escherichia coli polymerase, but no o subunit. The purified 2. antibioticus polymerafis als...

Respiratory chain of antimycin A-producing Streptomyces antibioticus.

An active respiratory chain system was demonstrated in sonically treated mycelium of Streptomyces antibioticus, the producer of antimycin A. The respiratory electron transfer from substrate to oxygen proceeded successively through flavoprotein(s), b-, c-, and a-type cytochromes, and terminated with the cyanide-sensitive cytochrome oxidase. The cytochrome composition of the culture was not affec...

Relationship between changes in the translational apparatus and actinomycin production in Streptomyces antibioticus.

As previously reported (G. H. Jones, 1975), transfer ribonucleic acids (tRNA's) and ribosomes from actinomycin-producing cultures of Streptomyces antibioticus show a decreased ability to function in aminoacylation and translation as compared with the corresponding components from younger cells. Further, specific changes in the isoacceptor patterns are revealed when tRNA's from actinomycin-produ...

Isolation and Identification of Streptomyces ramulosus from Soil and Determination of Antimicrobial Property of its Pigment

Background and objective: Pigment production by microorganisms is important due to more rapid growth, higher efficiency, easier extraction, and antimicrobial effects compared to other methods. The aim of the present study is to isolate and identify antibiotic pigment-producing actinomycetes from the soil of the Persian Gulf and to evaluate the antibiotic activity of their pigments in the second...
